Some say a best man is like a brother, but it is a whole different ball game when you are both the best man AND the brother.

The big different is that since you have known the groom (because he is your brother) since you were born (or he was), you have your whole life to get to know him – a whole life full of stories and experiences. This gives you the opportunity provide a very open speech.
And since you were most likely at home when your brother first brought his then girlfriend home, you probably have a much better grasp of the story and can share a little bit more.
The fact that your brother chose you to be his best man lets you know how much he values the relationship he has with you. So start with that and write a speech that matches the responsibility.
